Sample Collection - Chytrid Testing
Follow the guidelines below for step by step instructions to swab and send samples for Batrachochytrium dendrobatidis (Chytrid) testing.
- Using a plain swab (i.e. no transport medium), wipe the underside of the legs, feet and drink patch 3-5 times each, with the same swab tip.
- There is limited published evidence that fine tip rayon tipped swabs (e.g. Medical Wire & Equipment Co. MW 100–100) are most sensitive for DNA recovery (Boyle et al, 2007*). These are available through IZVG if required, and if the testing process can be delayed whilst these are ordered and sent out.
- For screening each frog, use one swab per frog. Identify each swab/frog separately.
- For screening a group, either use the same single plain swab on several frogs, or use a different swab tip on each frog, and then send all together in one pot per group (with a request for pool testing).
- Plain swabs should be sent by routine post at room temperature in green boxes or other packaging, to the usual Station House address.
- Use a standard IZVG Pathology aquarium or zoo submission form and write “Chytrid PCR test” in the OTHER box.
- It is advised to pack swab samples separately from formalin-fixed tissues, as formalin fumes/spillage can damage the nucleic acids on the swabs and reduce PCR sensitivity.
